Several new compounds now in the early clinical testing stage offer the hope of improving on the already impressive results shown with Avastin, Lucentis and related compounds. An interim analysis of 12- and 18-month data presented at the annual AAO conference concluded that fenretinide was well tolerated and showed a trend in favour of reducing lesion size in patients with AMD.
In another presentation, researchers reported that systemic immunosuppressive therapy with daclizumab and sirlolimus appeared to decrease retreatment rates in patients receiving anti-VEGF therapy.
Two other agents, alpha-5-beta-integrin (volociximab) and E10030 appeared safe and potentially effective in phase I studies. Researchers were particularly excited by preliminary results with a complement C3 inhibitor, POT-4. When given by the intravitreal route POT-4 was safe and well tolerated and showed sustained relief properties over a period of months, with improvements seen in chronic macular oedema.
Finally, VEGF-TRAP, an anti-VEGF agent, showed lasting improvements in best-corrected acuity in an 18-month phase II trial.
